Is there an API call to add one wrap-up code to multiple queues at a time? I've done a search but could not find anything.
Basically I need an efficient way to assign a new code to 50 queues.
There isn't a bulk update resource for queue wrapup code configuration; you'll need to update each queue individually. A simple loop over a data set in a script should work for this.
